ImmunoScape unlocks high-impact visibility into the immune system

Our proprietary multiplexed tetramer technology surpasses the limitations of flow cytometry and mass cytometry (CyTOF) for broader and deeper profiling of antigen-specific T-cells. Deep multiplexing is made possible by optimizing tetramers for high avidity TCR-antigen staining with combinatorial coding algorithms to produce unique metal barcodes. In a single sample, up to 120 antigen specificities can be interrogated, with several hundreds of parameters possible.

Together with our team, we offer expert analytical modelling and immunological data interpretation. Data generated through ImmunoScape’s immune profiling partnerships are securely stored and hosted by Amazon Web Services (AWS). TCR antigen profiling (TAP) to assess the antigen-specificity and repertoire

The initiative to establish TCR repertoire analysis in immunoSCAPE opens up new possibilities and the benefits are two fold (i) As a powerful tool,  it will allow us to obtain critical insights into the complexity of TCR repertoire of antigen-specific CD8+ T cells which will inform the design of future diagnostics strategy (ii) will add value to the existing resources at our disposal and provide immunoSCAPE with unique capability of extending these assays to tackle new biological problems.

TCR antigen profiling (TAP)

Our specific interest is to characterize the TCR repertoire of antigen-specific CD8+ T cells. Despite the advancements in studying cellular diversity and function, identification of target antigens remains a challenge. TCR antigen profiling (TAP) is a tool designed to address such important and challenging questions. At the heart of this initiative is the building of capacity to screen and detect antigen-specific CD8+ T cells and uncover their TCR repertoire. TAP technology will help us to maximize the utilization of precious clinical samples to obtain unique information.

At ImmunoSCAPE we have an established workflow for identifying T cell receptor and their cognate antigens by combining FACSorting and single cell sequencing capabilities.  Briefly CD8+ T cells are stained with oligo tagged MHC Class I and a panel of oligo tagged antibodies that are specific to cell surface proteins of interest. Antigen specific T cells are enriched by FACsorting. Subsequently the enriched T cells are subjected to single cell sequencing to obtain TCR, antigen and cell surface antibody specific readouts.
